Venosmil® [Hydrosmin]

Pharmacological properties

the active substance of venosmil is hydrosmin, according to its chemical structure it belongs to the group of flavonoids, it is 5.3’-mono-o- (β-hydroxyethyl) -diosmin and 5.3’-di-o- (β-hydroxyethyl) -diosmin. the mechanism of action of hydrosmin is not fully understood, but may be associated with the inhibition of catecholamine degradation, mainly by inhibiting catecholamine-o-methyltransferase. four levels of action of hydrosmin can be distinguished:

• a) the effect on permeability: reduces the permeability and fragility of capillaries (increase under the influence of histamine, bradykinin and unbalanced nutrition);
• b) the effect on the hemorheological properties of red blood cells: increases the ability of red blood cells to deform and reduces blood viscosity;
• c) effect on the tone of the veins: stimulates a constant and spontaneous reduction of smooth muscles of the venous wall;
• d) effect on the circulation of lymph: hydrosmin causes dilatation of the lymphatic vessels and increases the speed of the lymph, thereby improving lymph flow.

Venosmil stops the symptoms of chronic (peripheral) venous insufficiency (pain, feeling of heaviness, swelling). Hydrosmin has a pronounced effect on the manifestations of venous stasis caused by varicose dilatation of the vessels of the lower extremities.

Pharmacokinetics After a single oral administration of hydrosmin in healthy volunteers, it was noted that the curve of the concentration of the drug in the blood plasma in relation to time has a biphasic character. The initial peak in the concentration of the drug in blood plasma after use is observed after 15 minutes, after which the concentration begins to decrease slowly; 4 hours after application, a new increase in the level is observed with reaching an equilibrium concentration 5-8 hours after the use of the drug, after which the indicators decrease and after 24 hours after application they are practically not detected.

Within 48 hours, 90% of the dose of hydrosmin eliminated. Excretion of the active substance occurs mainly with feces (80%), 16–18% is excreted in the urine.

Indications

The drug is used for short-term relief (within 2-3 months) of edema and symptoms associated with chronic venous insufficiency (including hemorrhoids).

Application

The drug is taken orally during meals, washed down with water.

Adults are prescribed 1 capsule (200 mg) 3 times a day (every 8 hours), the duration of treatment is 3 months.

With exacerbation of hemorrhoids, 2 capsules (400 mg) are prescribed 3 times a day (every 8 hours), the duration of treatment is 3-4 weeks. Improvement occurs on the 2nd – 3rd week of treatment.

The use of the drug for 3 months with interruptions in treatment for 1–2 months for 1–2 years can be recommended for patients with chronic venous insufficiency or post-thrombotic syndrome.

Contraindications

Hypersensitivity to the active substance or other components of the drug.

Side effects

Typically, drug therapy is well tolerated. most frequent adverse reactions:

from the gastrointestinal tract: pain in the epigastric region, nausea;

on the part of the skin: cracks, itching;

from the central nervous system: weakness, headache.

special instructions

During pregnancy and breastfeeding. There are no clinical studies of the use of the drug during pregnancy. during preclinical studies, there was no direct or indirect negative effect on the course of pregnancy, fetal development. the use of the drug during pregnancy is possible only if the expected benefit to the mother outweighs the possible risk to the fetus. There is no data on the penetration of the drug into breast milk, so it is not recommended to use it during lactation.

Children. The effectiveness and safety of the drug in children have not been established.

It does not affect the reaction rate when driving vehicles or working with other mechanisms.

Interactions

Not described.special studies of the pharmacokinetics and pharmacodynamics of hydrosmin with simultaneous use with other drugs or food have not been carried out.

Overdose

Due to the good tolerance of venosmil, the possibility of intoxication is practically excluded, even in case of an overdose.
