Pharmacological properties
Sulfadimethoxine (4- (para-aminobenzenesulfamido) -2,6 - dimethoxypyrimidine) is a long-acting sulfonamide preparation. Sulfadimethoxine exhibits antibacterial activity against gram-negative and gram-positive bacteria: pneumococci, streptococci, staphylococci, Escherichia coli, Friedlender bacillus, dysentery pathogens; less active against protea. active against trachoma virus. Sulfadimethoxine does not affect strains of bacteria resistant to sulfonamides.
The drug is low toxic. When taken orally, it is absorbed relatively slowly. With a single dose in a therapeutic dose (1-2 g), the maximum concentration in the blood is reached after 3-12 hours. Maintenance doses (0.5-1 g) provide the necessary therapeutic concentration of the drug in the blood during the course of treatment. Compared with other long-acting sulfonamides, Sulfadimethoxine penetrates worse through the BBB (use for purulent meningitis is impractical). The drug is excreted in the form of metabolites, mainly with urine.
Indications
Diseases caused by microorganisms sensitive to sulfonamides: pneumonia, bronchitis, tonsillitis, otitis media, sinusitis, meningitis, diseases of the urinary and biliary tract, dysentery, pyoderma, gonorrhea, erysipelas, trachoma, wound infection, etc. are used in combination with resistant forms of malaria antimalarial drugs.
Application
Inside, 1 time per day in a daily dose with intervals between doses of 24 hours for mild and moderate forms of the disease, 1 g and 2 g are prescribed on the first day, respectively, on the following days, 0.5 g and 1 g for severe forms are used in combination with antibiotics. doses for children: on the 1st day - 25 mg per 1 kg of body weight, in the following days - 12.5 mg per 1 kg of body weight. for children over the age of 12, the initial and maintenance doses are 1 g and 0.5 g. After normalization of body temperature in maintenance doses, another 2-3 days are used. the duration of the course of treatment depends on the severity of the disease and is 7-14 days.
Contraindications
History of severe toxic allergic reactions to sulfonamides (agranulocytosis, hemolytic anemia, drug fever, severe dermatitis, hepatitis).
Side effects
Headache, dyspeptic symptoms, skin rash, drug fever, leukopenia. with the development of adverse reactions, the dose should be reduced or the drug should be discontinued.
