Pharmacological properties

promestrin - a synthetic estrogen-like product intended for topical use, has a local estrogenic effect on the mucous membrane of the lower parts of the urogenic tract, restores trophism. with intravaginal administration does not have a systemic effect, therefore, does not affect the endometrium, mammary glands and pituitary gland.

Promethrin has a local estrogenic effect on the vaginal mucosa: it promotes the proliferation of vaginal epithelium.

Pharmacokinetics With intravaginal administration, the drug interacts with the vaginal secretion, breaks down and its components are released. Promethrin is not cumulated in tissues, less than 1% of promethrin is absorbed, its T_½ is 24 hours. When administered intravaginally, promethrin does not have a resorptive effect, there are no systemic hormonal effects.

Indications

Vaginal atrophy due to estrogen deficiency. delayed healing of the vagina, cervix and vulva after childbirth, surgery, physiotherapy.

Application

Capsules 1 capsule (previously moistened) is administered deep into the vagina for 20 days. if necessary, treatment can be continued (menopause, castration, the use of estrogen-progestin-containing contraceptives).

Cream. Apply the cream 1-2 times a day every 2-3 days.

Children. Do not use.

Contraindications

Hypersensitivity to any component of the drug. despite the absence of systemic effects, this drug should not be used, like any estrogen, in estrogen-dependent tumors (such as breast cancer, endometrium).

The drug should not be used simultaneously with spermicides, latex condoms.

For a cream preparation, also: endometriosis (diagnosed or suspected).

Side effects

On the part of the skin and subcutaneous tissue: very rarely - local reactions, including irritation, itching, burning.

From the immune system: very rarely - allergic reactions.

The capsule preparation contains sodium methylparaben (E219) and sodium propylparaben (E217), which can cause allergic reactions (sometimes delayed).

Use during pregnancy or lactation. Colpotrophin is not indicated for use during pregnancy. However, the results of numerous epidemiological studies show that, unlike diethylstilbestrol, the use of estrogen in the early stages of pregnancy is not accompanied by a risk of malformations. When using a condom, there is a risk of rupture, because the oily substances and lubricants in capsules containing mineral oils reduce its strength.

Given the lack of sufficient data on the penetration of the drug into breast milk, its use during lactation should be avoided.

Interactions

The drug can inactivate the effect of any spermicidal agents, so their simultaneous use is not recommended.the drug should not be used when using a latex condom, since oily substances and lubricants containing mineral oils reduce its strength and increase the likelihood of rupture.

Overdose

Given the route of administration and low resorption of promethrin, systemic overdose is unlikely.

However, an overdose can be accompanied by an increase in local adverse reactions (irritation, itching, burning) in the vulva.

Therapy is symptomatic.
