The effect of the drug Cyclo 3 Fort on venous tone is dose-dependent with a single dose and is the result of the complex effect of all components of the drug.

The venotonic effect of the needle needle extract is due to direct stimulation of postsynaptic α-adrenergic receptors of smooth muscle cells of the vascular wall, as well as indirect effect due to the release of norepinephrine from granules of presynaptic nerve endings.

The intensity of the action of the needle extract is proportional to the temperature. In in vitro experiments on an isolated perfused vein, the needle extract quickly (after 5–8 min) caused a pronounced, progressive and prolonged contraction of the vessel. In animal experiments in vivo, needle extract induced an increase in venous pressure. The intensity of this effect in the case of healthy and pathologically altered veins was comparable.

The venotonic effect of the drug on human vessels is confirmed by the method of stereomicroscopic examination of the dorsal veins of the wrist.

The drug improves lymphatic circulation. In experiments on dogs, a significant and lasting increase in the speed of lymph flow in the thoracic lymphatic duct was revealed.

Angioprotective effect. The drug reduces the permeability of human capillaries (as demonstrated by the Landis test), and also increases the resistance of capillaries (as confirmed by induction of petechiae in the skin under vacuum). A significant increase in capillary resistance was noted already in the first hour after taking the dose. This effect is associated mainly with the action of ascorbic acid.

Pharmacokinetics Pharmacokinetic studies in animals have shown that needle extract (labeled with tritium isotope) and hesperidin methylhacone (labeled with carbon isotope) ¹⁴C) are rapidly absorbed. C_max of both ingredients in the blood plasma is reached after 2 hours. The components of the drug are excreted in urine and feces (enterohepatic recirculation is detected).

It is impossible to conduct appropriate studies of the pharmacokinetics of the drug in the human body, however, pharmacodynamic tests make it possible to indirectly evaluate the kinetics of the drug Cyclo 3 Fort. When stereomicroscopic studies of the dorsal veins of the wrist, the maximum venotonic effect was observed 2 hours after taking the drug with a return to the initial state after 6 hours.

Indications

Symptomatic treatment of venous-lymphatic insufficiency (with manifestations such as heaviness in the legs, pain, tired legs syndrome).

Chronic lymphatic insufficiency, including after injuries and surgical interventions, conditions after mastectomy.

Acyclic uterine bleeding caused by the use of microdose progestogen contraceptives or intrauterine devices after clinical and paraclinical examination of patients.

Exacerbation of hemorrhoids (treatment of functional symptoms).

Application

Inside, regardless of the meal. the capsules are swallowed whole and washed down with a glass of water. the duration of treatment is determined by the course of the disease and tolerance to the drug.

The usual dose for adults is 2-3 capsules per day.

Doses for patients with proctologic diseases - 4-5 capsules per day for the first 3 days, then 2 capsules per day for 4 days.

Contraindications

Hypersensitivity to the components of the drug, severe kidney disease.

Side effects

From the gastrointestinal tract: diarrhea, sometimes severe (there is a risk of weight loss and impaired water / electrolyte balance), quickly passing after discontinuation of the drug; nausea, stomach pain.in cases where the drug was not canceled with the onset of diarrhea, microscopic colitis, mainly lymphocytic, rarely developed.

From the immune system: allergic reactions, including skin rash, Quinckes edema, urticaria; sometimes - anaphylactic shock in the presence of sensitization.

Application for hemorrhoids: treatment with Cyclo 3 Fort cannot be a substitute for specific treatment of proctologic diseases. Cyclo 3 Fort treatment should be short. If the symptoms do not quickly disappear, it is necessary to conduct a thorough proctological examination of the patient and prescribe the appropriate treatment.

If you experience nausea or stomach pain, the drug should be taken with food.

When taken in high doses and prolonged use of ascorbic acid, it is necessary to control renal function and blood pressure, as well as pancreatic function. Use the drug with caution in patients with a history of kidney disease or diabetes.

In urolithiasis, the daily dose of ascorbic acid should not exceed 1 g. The drug should not be prescribed in high doses to patients with increased blood coagulation.

Since ascorbic acid increases iron absorption, its use in high doses can be dangerous for patients with hemochromatosis, thalassemia, polycythemia, leukemia and sideroblastic anemia.

Patients with a high iron content in the body should use the drug in minimal doses. Absorption of ascorbic acid may be impaired with intestinal dyskinesias, enteritis and achillia. It is used with caution in patients with glucose-6-phosphate dehydrogenase deficiency. As a reducing agent, ascorbic acid can affect the results of laboratory tests, for example, when determining the blood glucose, bilirubin, transaminase activity, LDH, etc. Since ascorbic acid has a mild stimulating effect, it is not recommended to take the drug at the end of the day.

During pregnancy and breastfeeding. Clinical observations did not reveal the fetotoxic effect of the Cyclo 3 Fort drug, so it can be used during pregnancy.

Since there is no information on the allocation of the drug Cyclo 3 Fort with breast milk, the drug should not be prescribed to women who are breast-feeding.

Children. Do not use.

Interactions

Specific studies of the interaction of the drug cyclo 3 fort with other drugs or food products have not been conducted.

The absorption of ascorbic acid is reduced with the simultaneous use of oral contraceptives, the use of fruit or vegetable juices, alkaline drinking. Oral ascorbic acid increases the absorption of penicillin, tetracycline, iron, reduces the effectiveness of heparin and indirect anticoagulants, and increases the risk of crystalluria during treatment with salicylates. The simultaneous intake of vitamin C and deferoxamine increases the tissue toxicity of iron, especially in the heart muscle, which can lead to decompensation of the circulatory system. Vitamin C can be taken only 2 hours after injection of deferoxamine. Long-term use in high doses by persons taking disulfiramine inhibits the disulfiram-alcohol reaction. High doses of the drug reduce the effectiveness of tricyclic antidepressants, antipsychotics - phenothiazine derivatives, tubular reabsorption of amphetamine, disrupt the excretion of mexiletine by the kidneys. Ascorbic acid increases the overall clearance of ethyl alcohol.Quinoline drugs, calcium chloride, salicylates, glucocorticoids with prolonged use reduce the reserves of ascorbic acid in the body.

Overdose

Cases of overdose are not registered. an increase in the severity of adverse reactions is possible. when using ascorbic acid at a dose of 1 g / day, irritation of the mucous membrane of the digestive tract, heartburn, nausea, vomiting, diarrhea are possible.

From the urinary system: with prolonged use of ascorbic acid in high doses, damage to the glomerular apparatus of the kidneys, crystalluria, the formation of urate, cystine and / or oxalate calculi in the kidneys and urinary tract are possible.

Treatment: symptomatic therapy.
